چکیده
Receptor-mediated drug delivery is a Trojan horse approach for the release of therapeutics into cells, tissues, or organs. By coupling drugs to ligands of cell surface receptors, ligand-drug conjugates can accumulate in specific tissues and can be internalized by cells through the mechanism of receptor-mediated endocytosis.1 Receptors targeted in this way include the low-density lipoprotein (LDL) receptor,2 transferrin receptors,3 and folate receptors.4 Receptors involved in the passage of nutrients across the blood-brain barrier through the related mechanism of receptor-mediated transcytosis have been similarly targeted to deliver drugs and imaging agents into the brain.5 This latter strategy is of significant interest because less than 2% of all drugs are capable of penetrating the bloodbrain barrier.5 We report here a method for synthetic receptor-mediated drug delivery. This approach was applied to the glycopeptide antibiotic vancomycin (1), a drug of last resort against bacterial pathogens

Receptor-mediated endocytosis is a highly efficient mechanism for cellular uptake of membrane-impermeant ligands. Cells use this process to acquire nutrients, initiate signal transduction, promote development, regulate neurotransmission, and maintain homeostasis. Natural receptors that participate in receptor-mediated endocytosis are structurally diverse, ranging from large transmembrane protei...
متن کاملSynthetic mimics of mammalian cell surface receptors: prosthetic molecules that augment living cells.
Specific receptors on the surface of mammalian cells actively internalize cell-impermeable ligands by receptor-mediated endocytosis. To mimic these internalizing receptors, my laboratory is studying artificial cell surface receptors that comprise N-alkyl derivatives of 3beta-cholesterylamine linked to motifs that bind cell-impermeable ligands. When added to living mammalian cells, these synthet...
متن کاملIn Silico Design and Verification of LAMP-BDNF Chimeric Protein for Presentation of BDNF on the Surface of Exosomes for Drug Delivery Through Blood-Brain Barrier
Background and purpose: The mature form of brain-derived neurotrophic factor (BDNF) binds to BDNF/NT-3 growth factors receptor (Trk-B). This binding leads to activation of Ras–MAPK pathway which is integrated with cell growth and proliferation. The BDNF deficiency is correlated with various diseases and affects aging and miscellaneous. In the present study we aimed to design a chimeric LAMP-BDN...
